Several years ago, I moved a production SendMail system that supported 500+ users from a Sun SPAR environment to a Linux PC, with barely a hiccup. Linux has a reputation for being tailor made for Web and e-mail servers. The number you mention should be more than sufficient.
Make sure you get the processor, memory and disk you need for your environment, and you should be good to go. Don't modify sendmail.cf with linuxconf; you could run into problems. Obviously the administration tools are different as far as the OS is concerned -- which may be your biggest challenge -- but overall SendMail is SendMail. If you know how to run an e-mail system, you should be fine.
